An Optimized Protocol for Indirect Organogenesis from Root Explants of Agapanthus praecox subsp. orientalis ‘Big Blue’

Agapanthus praecox has become a burgeoning variety in the flower market due to its high ornamental value with unique large blue-purple inflorescence. For rapid entering into market, tissue culture technology or organogenesis an attractive application over conventional reproduction approach. In this study, highly efficient protocol based on indirect been successfully established for A. subsp. orientalis ‘Big Blue’. Two types of explants, root tips versus segments, were compared callus induction frequency response media. The media contain Murashige and Skoog’s (MS) Basal Salt supplemented various concentrations picloram (PIC), 2,4-Dichlorophenoxyacetic acid (2,4-D), thidiazuron (TDZ), kinetin (KT) naphthalene acetic (NAA). Of two found be more effective than segments. Among tested, highest rate (100.00%) was achieved when cultured MS 2.0 mg/L PIC, 1.5 KT 0.1 NAA, which probably accredited higher endogenous phytohormone contents, especially 3-indoleacetic (IAA). optimal medium proliferation + 1.0 PIC 6-BA 0.4 fresh weight increased by 72.74%. After being transferred onto adventitious bud 25 days, shoots dedifferentiated from surface flourishing callus, then developed plantlet roots 90 days. plantlets transplanted greenhouse survival 92.86%. This study innovatively system as provided practical reference application.
